Bovineplasmaamineoxidasehasamolecularweightof170kDaandanoptimumpHof6.2forspermineand7.2forspermidine.Amineoxidasesaredividedintotwoclasses:thepyridoxalandcoppercontainingenzymetowhichplasmaamineoxidasebelongs,andtheFAD-containingaminooxidases.Naturalsubstratesincludecatecholamines,tryptaminederivativesandotherphysiologicallyactiveamines.Plasmaamineoxidaseisusedinresearchrequiringnitrogengrouptransfers.Themoleculeiscomposedoftwoidenticalpolypeptidechains.TherearetwopyridoxalphosphatesandtwoatomsofCu+permolecule.Bovineplasmaamineoxidaseisinhibitedbycopperchelatingagents,manycarboxylreagentssuchascuprizone,hydroxylamineandcyanide.Benzoicacidandbenzylalcoholarebothnon-competitiveinhibitors(KI=30and34mMrespectively).TheassayfordeterminationofamineoxidasesemployedatWorthingtonisessentiallythatofTaboretal.,JBC,208,645,(1954)withthereactiontemperaturereducedto25°C.
