AspergillusoryzaeRibonucleaseT2isamemberoftheRNaseT2familyofendonucleasesthatarepresentinawidevarietyofmicrobial,plantandanimalspecies.IncontrasttoAspergillusoryzaeRibonucleaseT1,whichisanexclusivelyguanylic-acidspecificendonuclease,allRNaseT2-likeenzymesareessentiallybasenon-specific.However,RNaseT2endonucleasesfromdifferentspeciescanshowslightbasepreferences.Thefungalenzymes,includingAspargillusoryzaeRNaseT2,showslightbasepreferenceinthefollowingorder:A>G>C,U.RNaseT2cleavesbetweenthe3’-phosphateresidueofonebaseandthe5’-OHresidueoftheadjacentnucleotideforminga2’,3’-cyclicphosphateintermediatefollowedbythegenerationofoligonucleotideswith3’-phosphateresidues.RNaseT2hasamolecularweightof36kDaand12-15%ofitsmassiscomposedofcarbohydrate.Ithasanisoelectricpointof5.0andoptimumactivityatpH4.5. RNaseT2isstronglyinhibitedbyCu++,Zn++andHg++andtoalesserdegreebyCa++,Mg++andheparin. MononucleotidesandRNaseT2digestionproductscanalsoactascompetitiveinhibitors.EDTAwillstimulateactivity,especiallyinthepresenceofdivalentcations.
