Trypsinisapancreaticserineproteasewithsubstratespecificitybaseduponpositivelychargedlysineandargininesidechains. Itisderivedfroma34kDainactiveprecursorzymogen,trypsinogen,afterenzymaticremovalofanN-terminal6-aminoacidleadersequenceresultinginthe23.8kDatrypsinmolecule. TheoptimumpHis8.0. Trypsinisinhibitedbyorganophosphoruscompoundssuchasdiisopropylfluorophosphateandnaturalinhibitorsfrompancreas. Soybean,limabean,andeggwhitearealsosourcesofnaturalinhibitors. TrypsincleavesamideandesterbondsofArgandLys. TheWorthingtonSequencingGradeTrypsinhasbeenfurtherpurifiedtoremovetracecontaminatingproteasesandautolysisproductswhichcouldinterfereintrypsindigestionexperiments,andexhibitsasinglebandonPAGE.
