Trypsinisapancreaticserineproteasewithsubstratespecificitybaseduponpositivelychargedlysineandargininesidechains.Itisderivedfroma34kDainactiveprecursorzymogen,trypsinogen,afterenzymaticremovalofanN-terminal6-aminoacidleadersequenceresultinginthe23.8kDatrypsinmolecule.TheoptimumpHis8.0.Trypsinisinhibitedbyorganophosphoruscompoundssuchasdiisopropylfluorophosphateandnaturalinhibitorsfrompancreas.Soybean,limabean,andeggwhitearealsosourcesofnaturalinhibitors.TrypsincleavesamideandesterbondsofArgandLys.TheWorthingtonSequencingGradeTrypsinhasbeenfurtherpurifiedtoremovetracecontaminatingproteasesandautolysisproductswhichcouldinterfereintrypsindigestionexperiments,andexhibitsasinglebandonPAGE.
